Isolation and Purification of Proteases by Immobilization of Protease Inhibitors

Marimastat is an inhibitor of the hydroxamate type for matrixmetalloproteinases showing practically no selectivity for any distinct member of this protein family. A new, optimised synthesis route was developed for Marimastat by which a suitable linker molecule can be attached. The synthesised conjugates are applied for experiments with surface plasmon resonance (SPR) and for affinity chromatographic purification of MMP from cellular and tissue extracts.
